However would it not be: What I like most it is your smile

Why is c'est -> is 

It should be

c'est -> it is

Your remark is correct, in that in French one often repeats the subject. It's like saying in English: "What I like best, that is your smile." You could omit the "that" in English, but in French it is common to say it.
